At present, except that Bacillus coagulans can withstand 100 ℃ high temperature in the market, most probiotics are suitable for storage at about 2 ℃ – 10 ℃ at room temperature, and the maximum storage temperature is no more than 50 ℃, otherwise probiotics will be almost wiped out. The maximum temperature difference between North and South in China is about 70 ℃ and that between winter and summer is about 45 ℃, which poses a very high challenge to the transportation and storage of probiotics. Probiotic products will undergo many changes in environment and temperature from the completion of production to the final arrival in the hands of consumers, which also leads to the loss of probiotics.

Threshold 3: complex human structure leads to non combat reduction of probiotics
As we all know, the human body has a powerful digestive system, which will produce some highly corrosive secretions to digest and decompose food. In order to survive in the intestinal mucosa, microorganisms must endure many extreme environments. Changes in pH, hypoxic levels, nutritional limitations and elevated osmotic pressure all constitute potential obstacles to survival.
In the process of eating probiotics, probiotics enter the gastrointestinal tract after eating through the mouth. Only after the invasion of gastric acid and choline in the digestive tract can they reach the intestinal tract smoothly and colonize the intestinal tract to play its role. Generally, most of them are killed in the stomach (Ph1 ~ 2) and can survive and play a role only after entering the small intestine (ph6 ~ 8).
In the experiment of simulating gastric juice environment, it shows that the longer the probiotics stay in gastric juice, the lower the survival rate, showing a trend of accelerated attenuation.
At the same time, the liver secretes up to one liter of bile into the intestine every day, and contacting bile is also a serious challenge. Bile is the secretion of digestive tract, which plays an important role in the emulsification and solubilization of lipids. It can affect the phospholipids and proteins of cell membrane and destroy the homeostasis of intracellular environment. Therefore, the ability of pathogens and commensals to tolerate bile is important for their survival and subsequent colonization in the intestinal mucosa.
